The last few decades have seen a significant increase in certain types of cancers and a shift toward genes as a cause of the disease.
There have been many claims of genetic cancers, but treatments based on this idea are lacking.
Recently, researchers investigating a genetic mutation alleged to cause breast, ovarian, colorectal, and lung cancer…
…discovered that aspirin is more effective than anything they came up with in decades of genetic research.
The cell and animal research was carried out at the Beth Israel Deaconess Medical Center in Boston, Massachusetts. This paper was published in Cancer Research.
Years ago, researchers discovered that certain types of cancer are correlated with genetic mutations in PIK3CA.
These mutations are commonly found in various cancers, including breast, ovarian, colorectal, and lung cancer.
But all attempts to treat these cancers by inhibiting this mutation have had miserable results.
“Despite the high incidence of oncogenic mutations in PIK3CA. PI3KCA inhibitors have yielded little clinical benefit for breast cancer patients.”
However, numerous reports in the research literature note that aspirin seems to help with cancers that correlate with this mutation.
“Recent epidemiologic studies have suggested a therapeutic benefit from aspirin intake in cancers harboring oncogenic PIK3CA.”
Researchers first carried out some cell experiments.
In these experiments, they compared aspirin’s ant-icancer effect on normal cells against its effect in cells with these genetic mutations.
Aspirin has well-known anti-cancer effects.
These experiments showed that the anti-cancer effect of aspirin was even more potent in cells with this mutation.
“Here, we show that mutant PIK3CA-expressing breast cancer cells have greater sensitivity to aspirin-mediated growth suppression than their wild-type counterparts.”
The researchers looked at the anchorage-independent growth of the cells.
This is the ability of these cells to grow and multiply, forming tumors without the need to be attached to any surface.
This “anchorage-independent growth” may help cancer spread around the body more rapidly.
Aspirin reduces the growth of free-floating cancer cells with this mutation.
“Aspirin decreased viability and anchorage-independent growth of mutant PIK3CA breast cancer cells.”
The researchers identified 3 potential anticancer mechanisms of aspirin in this context.
These effects include activating AMPK, an enzyme with multiple roles in cancer, and decreasing mTORC1.
mTORC1 is part of the mTOR group of proteins/enzymes, which is being heavily researched in aging and diseases of access growth – like cancer.
mTOR-inhibiting treatments like rapamycin are very promising anti-cancer, anti-aging compounds.
Aspirin is an mTOR inhibitor like rapamycin.
Aspirin also increases autophagy, the process of destroying and recycling cell materials.
Increasing autophagy can suppress tumor growth by removing damaged organelles and proteins.
“We ascribed the effects of aspirin to AMPK activation, mTORC1 inhibition, and autophagy induction.”
The researchers then carried out several animal experiments.
They found that aspirin reduces the growth of breast cancers in mice with this mutation in rodents.
“PIK3CA-driven mouse mammary tumors treated daily with aspirin resulted in decreased tumor growth. Combination therapy of aspirin and a PI3K inhibitor further attenuated tumor growth.”
The amount of effective treatments for cancer generated by the idea of genetically determined cancers…
…is shockingly low compared to the amount of research showing that simple, old aspirin is a potent anti-cancer substance.
“Our study supports the evaluation of aspirin and PI3K pathway inhibitors as a combination therapy for targeting breast cancer.”
Aspirin is long out of patent as it was first synthesized in 1897.
